Obtaining Transgenic Elite Indica Restorer Line and Study on Genetic Stability of Transgenes

转抗虫基因优良籼型恢复系的获得及其外源基因的遗传稳定性研究

Abstract: The transgenic elite indica restorer line containing snowdrop lectin(Galanthus nivalis agglutinin, GNA) gene gna was obtained from elite indica restorer line Shuhui 527 by biolistics. Results indicated by PCR, PCR Southern blotting and Southern blotting showed that the transgenes were transmitted from the first generation (T0) of transgenic plants to the third generation(T2). The plant height and seed setting rate of the first generation (T0) of transgenic plants were reduced apparently as compared with the corresponding plants derived from seeds and untransformed calli. However, this kind of variation could not be inherited stably. The usage of mini preparation DNA in selection of transgenic plants was also discussed.

摘要: 用基因枪法将雪花莲外源凝集素基因(gna)转移到优良籼型杂交稻恢复系蜀恢527中,通过PCR、PCR Southern blotting和Southern blotting等分子方法检测证明,外源基因已稳定遗传到转基因第三代(T2)。转基因第一代植株(T0)在株高和结实率上与相应的组培、种子实生苗植株相比,发生明显的不可遗传的变异。随着繁殖代数的增加,转基因植株恢复到与对照植株一致。还讨论了DNA微量提取法在转基因后代筛选中的运用。
