水稻细菌性条斑病种子带菌检测技术研究:I. 免疫放射分析法

Abstract: Immunoradiometric assay (IRMA) is a technique of superior trace analysis which integrated the specificity of serology and sensitivity of radioisotope. With the method in rice seed inspection of X. c. pv. oryzicola for 3 years. 402 seed samples were tested. The results showed that equal or above 1000 bacterial cells per ml of X.c. pv. oryicola in the seeds could be detected with the IRMA. The antiserum and labelled protein A was stable and only slight cross-reaction with X. c. pv. oryzae. The process of seed inspection for a batch of seeds could be finished in 4-5 hours if the antiserum and labelled protein A was prepared Compared with the concentration inoculation of common methods, the IRMA was more effective and reliable.

摘要: 免疫放射分析法(IRMA)集放射性同位素的灵敏性与抗原-抗体反应的特异性为一体。该方法应用于水稻细菌性条斑病种子带菌检测,经三年试验表明这一方法具有灵敏度高(最小检测浓度为10^3细菌/ml)。特异性强,稳定性好。快速(4~5小时内可得出结果)。通过402份样品的检测,经与常规的浓缩接种法比较,表明该方法应用于带菌稻种的检测前景广阔。
