Detection of Plasmid pCAMBIA 1301 by DNAchip

Abstract: DNAchip was used to detect plasmid pCAMBIA 1301 in transgenic rice. To be different to biochip based on hybridization, minisequencing biochip was used to detect report gene hptⅡ, aadA, Gus and promoter CaMV 35S in plasmid pCAMBIA 1301. The minisequencing biochip depends on both hybridization and DNA polymerase to detect sample. Asymmetric PCR was better than normal PCR in sample preparation.

摘要: 探讨了利用基因芯片对转基因水稻中质粒pCAMBIA 1301的检测。不同于以往基于杂交检测基础上的基因芯片,利用微测序芯片实现了对质粒pCAMBIA 1301中报告基因hptⅡ、aadA、Gus,启动子CaMV 35S的检测。这种微测序芯片不仅通过杂交检测,还利用DNA聚合酶的特异性来实现对样品的检测。对样品进行常规PCR和不对称PCR处理实验表明,经不对称PCR处理后样品的检测效果明显优于常规PCR处理。
