cDNA Cloning and Sequence Analysis of Genome Segment S10 from Rice Black-Streaked Dwarf Virus

水稻黑条矮缩病毒基因组片段S10的cDNA克隆及全序列分析

Abstract: Genome segment S10 of a Zhejiang isolate of rice black-streaked dwarf virus (RBSDV-Zj) was amplified by RT-PCR and sequenced. It consisted of 1801 nt (EMBL accession number is AJ297433) with only one open reading frame which encodes a polypeptide with molecular weight of 63.1 kD. Sequence comparison showed 93.8% and 96.2% homology at level of nucleotide and amino acid sequences between RBSDV-Zj and Japanese isolate (RBSDV-Jap), respectively;and shared 87.1% and 92.0% homology at level of nucleotide and acid sequences between RBSDV-Zj and Italian MRDV, respectively.

摘要: 基于RT-PCR策略克隆了水稻黑条矮缩病毒(rice black streaked dwarf virus,RBSDV)浙江分离株基因组S10,并测定了它的全序列。结果表明,水稻黑条矮缩病毒浙江分离物(RBSDV-Zj)基因组片段S10全长1801 nt(EMBL登录号为AJ297433),仅含有一个大的开放阅读框(open reading frame,ORF),编码一个63.1 kD的多肽,与日本的水稻黑条矮缩病毒基因组片段S10在核苷酸和氨基酸水平上同源性分别为93.8%和96.2%;与意大利玉米粗缩病毒(maize rough dwarf virus,MRDV)的基因组片段S10在核苷酸和氨基酸水平上的同源性分别为87.7%和92.0%,推测它们是同一病毒的不同地理小种。
